AAS Trust Travel Grant Awardee Report

I received an AAS Trust Travel Grant, which contributed significantly towards my travel expenses to the Arteriosclerosis, Thrombosis and Vascular Biology (ATVB) Annual Conference 2008 held April 16-18, 2008 in Atlanta, GA.

 

I presented two posters at the ATVB Conference. The first poster was entitled “Dose-dependent effect of rosuvastatin on very-low density lipoprotein apolipoprotein C-III kinetics in men with the metabolic syndrome”. The study presented resulted from collaborations with AstraZeneca (Australia and United Kingdom) and Professor Paul Nestel at the Baker Heart Research Institute, Melbourne. This study investigated the dose-dependent effect of rosuvastatin on very-low density lipoprotein (VLDL) apoC-III kinetics in men with the metabolic syndrome in a randomized, double blind, three-way crossover placebo-controlled clinical trial. The kinetics of VLDL apoC-III were measured using a stable isotope (D3-leucine), gas-chromatography mass spectrometry and multi-compartmental modelling. The data suggest that rosuvastatin dose-dependently decreased VLDL apoC-III concentration by a mechanism related to an increase in VLDL apoC-III fractional catabolic rate (FCR) and a decrease in VLDL apoC-III production rate (PR). These findings provide a mechanistic explanation for the triglyceride-lowering effects of rosuvastatin with implications for cardiovascular risk reduction in the metabolic syndrome. This poster was short listed and won an ATVB Conference Merit Award for Young Investigators/New Investigator Travel Award. 

 

The second poster was entitled “Regulatory effect of atorvastatin and fenofibrate on apolipoprotein C-III kinetics in the metabolic syndrome”. The study investigated the effect of two different lipid-lowering agents, atorvastatin and fenofibrate on VLDL apoC-III kinetics in men with the metabolic syndrome. The data suggest that atorvastatin and fenofibrate decreased VLDL apoC-III concentration as a consequence of increased VLDL apoC-III FCR and decreased VLDL apoC-III PR. These findings may explain one of the lipoprotein-mediated benefits of these agents on triglyceride concentrations and hence, cardiovascular risk in the metabolic syndrome. 

 

In addition, I presented a talk at the Lipoprotein Kinetics Satellite Meeting on April 15, 2008 in Atlanta. The talk was entitled “Dose-dependent effect of rosuvastatin on very-low density lipoprotein apolipoprotein C-III kinetics in men with the metabolic syndrome”. The Kinetics Satellite Meeting acts as a platform to discuss methodological issues related to the performance of metabolic studies and the application of compartmental modelling to better understand the physiology and pathophysiology of complex systems.

 

The posters and oral presentations were very well received, and I obtained valuable feedback and comments from experts present at the conference. In addition, I met with Professor Ernst Schaefer (Tufts University) to discuss our collaborative work on a recently started systems biology project. I also met with Professors Murray Huff (University of Western Ontario) and Elizabeth Parks (University of Texas, Southwestern Medical Center), both who continue to provide informative insights into methodologies associated with performance of complex metabolic studies. I also had the pleasure of meeting with Professor Larry Rudel (Wake Forest University) who took significant interest in discussing the findings of my first poster and suggested potential collaborative work between his laboratory and our Metabolic Research Centre. 

 

I would like to thank the Australian Atherosclerosis Society for the funding support provided. Having the opportunity to apply one’s knowledge and to be challenged and tested in an international research arena further reinforces and substantiates ones’ desire to progress in research and innovation.

Cholesterol fails to break the ignorance barrier

DATA GATHERED for the recent National Cholesterol Awareness Campaign for the Australian Atherosclerosis Society reveals that 74 per cent of Australians with a family history of heart disease have not discussed cholesterol with their doctor in the past 12 months.

The National Cholesterol Test survey of more than 500 men and women aged between 18 and 55, conducted for the campaign by the Society ’s Familial Hypercholesterolemia Sub-Committee, indicated that: though heart disease is the leading cause of death in Australia, only six per cent of Australians had a cholesterol test because they were concerned about the disease, though heart disease causes more deaths than cancer each year, 48 per cent believed that cancer was the more concerning issue, more than 50 per cent believed that cholesterol mainly came solely from food, though most of the cholesterol it needs is made by the body itself, and 47 per cent actively tried to manage their cholesterol through lifestyle changes such as diet and exercise, not knowing that these may not always be enough to lower cholesterol to acceptable levels.
